Donatello has a need to know everything, which sometimes gets out of hand. By that I don't mean that he's really nosy or anything, he just sits a little too long in that lab of his some times, researching... he was of course the one who was most devoted in finding their origin.
Don is not very fond of violence, which shows in his choice of weapon, a simple "wooden stick".
He is often the one who has to retain peace among his brothers, and seldom fights with them himself. (outside of training, that is) He's an excellent ninja, but just more defensive.
He is kind of a loner like Raph, but prefers to be with his brothers instead of being locked up in his lab, unless he's working on something really important. He is also "Mr. Fixit", and the turtles would've been in trouble if he weren't there to mend broken things.
Don likes to participate in a good joke, but is known to make jokes that go over his brothers heads.
If he does have any negative thoughts about other people, he mostly keeps them to himself.
He is seen fighting once in a while with Raph, though, who calls him "nerd" and such when he retires to his laboratory...err, room.

What I think: Don is also one of my favorites, in another way than Raph. Don appeals to my intellectual side, (Do I have any? ;P) and he is definitely someone I'd like to discuss/talk with. OK, I also think he's incredibly cute! You've got me! Just not as "sexy" as Raph.
I would feel a little stupid compared to Don, but he's a really nice guy, and wouldn't be one to patronize.
I wonder if female attention is what it takes to get him out from that lab once in a blue moon?
It's uncertain how he would react if a girl liked him, because this has never been portrayed in neither the movies, comics nor in the cartoons. (At least nothing I know of)
Then, of course, there's always the hated/loved theory of him being gay...which I am, as you might understand, not particularly fond of, even if Phish's portration actually is an incredibly good one. Her Don's truly a strong character.
